產品說明
Violet Red Bile Glucose Agar（紫紅膽鹽葡萄糖瓊脂，VRBGA）是檢測與計數腸桿菌科細菌的標準選擇性培養基，ISO 21528 指定使用。膽鹽與結晶紫抑制革蘭氏陽性菌，以葡萄糖取代乳糖，所有腸桿菌科都能發酵而呈現紅紫色菌落並帶沉澱暈，涵蓋沙門氏菌等不發酵乳糖的菌屬，比只看乳糖的 VRBA 更完整。用於食品、乳品與環境衛生指標菌檢驗。
原理
Violet Red Bile Glucose Agar is a modification of Violet Red Bile Agar in which lactose is replaced by glucose, so that all members of the Enterobacteriaceae, including lactose-negative organisms, are detected.¹ Enzymatic digest of animal tissues and yeast extract supply nitrogen, amino acids, vitamins, and growth factors, and sodium chloride maintains the osmotic equilibrium. Bile salts and crystal violet inhibit Gram-positive bacteria. Glucose is fermented by all Enterobacteriaceae, producing acid that is indicated by neutral red. Characteristic colonies are pink to red or purple, with or without a zone of precipitated bile. The overlay restricts spreading and further suppresses non-target flora. Characteristic colonies are counted and confirmed by the oxidase test and glucose fermentation.²
選擇性培養基的檢驗流程
病原菌檢驗通常分三段：先用增菌培養基（如緩衝蛋白腖水）讓受損菌體復原並增加數量，再用選擇性增菌培養基抑制雜菌，最後用選擇性鑑別培養基分離出目標菌落。選擇性成分多為膽鹽、染劑或抗生素，用來抑制非目標菌；鑑別成分則讓目標菌長出特定顏色或型態的菌落。請依您採用的檢驗方法選擇對應的培養基組合。
配製與使用
Suspend 41.53 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. DO NOT AUTOCLAVE. Cool to 45-50°C and use for the pour-plate technique with an overlay of the same medium. Test the finished medium for performance using stable, standard control cultures.
