產品說明
Violet Red Bile Agar（紫紅膽鹽瓊脂，VRBA）是食品與乳品大腸桿菌群計數的標準選擇性培養基。膽鹽與結晶紫抑制革蘭氏陽性菌，發酵乳糖的大腸桿菌群產酸，使中性紅變色而形成紅紫色菌落並帶膽鹽沉澱暈。用於傾注平板法的大腸桿菌群計數，是乳品、飲料與食品工廠衛生指標檢驗的基本耗材。
原理
Violet Red Bile Agar is a selective and differential medium for the enumeration of coliforms by the pour-plate technique.¹ Enzymatic digest of animal tissues and yeast extract supply nitrogen, amino acids, vitamins, and growth factors, and sodium chloride maintains the osmotic equilibrium. Bile salts and crystal violet inhibit Gram-positive bacteria. Lactose is the fermentable carbohydrate, and neutral red serves as the pH indicator. Coliforms ferment lactose and produce acid, forming purplish-red colonies at least 0.5 mm in diameter, often surrounded by a reddish zone of precipitated bile. The overlay restricts spreading and creates semi-anaerobic conditions that further suppress non-coliform flora. Characteristic colonies are counted and, where required, confirmed in a lactose fermentation broth.²
選擇性培養基的檢驗流程
病原菌檢驗通常分三段：先用增菌培養基（如緩衝蛋白腖水）讓受損菌體復原並增加數量，再用選擇性增菌培養基抑制雜菌，最後用選擇性鑑別培養基分離出目標菌落。選擇性成分多為膽鹽、染劑或抗生素，用來抑制非目標菌；鑑別成分則讓目標菌長出特定顏色或型態的菌落。請依您採用的檢驗方法選擇對應的培養基組合。
配製與使用
Suspend 41.53 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. DO NOT AUTOCLAVE. Cool to 45-50°C and use for the pour-plate technique with an overlay of the same medium. Test the finished medium for performance using stable, standard control cultures.
