A-1 Broth 是液體培養基，屬於食品與臨床檢驗用的選擇性培養基，以選擇性成分抑制雜菌、以鑑別成分辨識目標菌落。用於水與貝類的糞便大腸桿菌群快速檢驗。上鼎以食品級原料製造，生產環境通過 ISO 22000 認證，每批附 COA。
原理
A-1 Broth was developed by Andrews and Presnell for the rapid recovery of fecal coliforms from estuarine water, eliminating the need for a presumptive enrichment step before confirmation.¹ Tryptone supplies nitrogen, amino acids, and peptides, while lactose and salicin serve as fermentable carbohydrates. Sodium chloride maintains the osmotic balance. Triton X-100, a non-ionic surfactant, suppresses the growth of non-fecal organisms and assists the recovery of stressed coliforms from marine samples. Tubes are incubated at 35°C for 3 hours to allow resuscitation of injured cells, and then transferred to a water bath at 44.5°C for a further 21 hours. At the elevated temperature, only thermotolerant (fecal) coliforms ferment lactose with the production of gas. The presence of gas in the Durham tube after incubation constitutes a positive result, and fecal coliform densities are calculated from the pattern of positive tubes using most probable number tables.²
選擇性培養基的檢驗流程
病原菌檢驗通常分三段：先用增菌培養基（如緩衝蛋白腖水）讓受損菌體復原並增加數量，再用選擇性增菌培養基抑制雜菌，最後用選擇性鑑別培養基分離出目標菌落。選擇性成分多為膽鹽、染劑或抗生素，用來抑制非目標菌；鑑別成分則讓目標菌長出特定顏色或型態的菌落。請依您採用的檢驗方法選擇對應的培養基組合。
配製與使用
Suspend 31.5 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Dispense into tubes containing inverted Durham tubes. Sterilize by autoclaving at 121°C for 10 minutes. Test the finished medium for performance using stable, standard control cultures.

食品級與實驗室級的差別

01選擇性培養基可以高壓滅菌嗎？
不一定。部分含有染劑或選擇性抑制劑的培養基過度加熱會失去選擇性，只需加熱至完全溶解、不可高壓滅菌。請依本產品的配製說明操作，不確定時歡迎聯絡我們確認。
