Buffered Listeria Enrichment Broth Base（緩衝李斯特菌增菌液基礎，BLEB）是美國 FDA BAM 與台灣食藥署「食品中單核球增多性李斯特菌之檢驗」指定的增菌液：以胰化酪蛋白大豆培養液為底，加酵母抽出物與磷酸鹽緩衝、丙酮酸鈉幫助受損菌體復原，增菌 4 小時後再依方法加入吖啶黃、萘啶酸與環己醯亞胺等選擇性補充物，30°C 培養 24 至 48 小時後取樣塗抹選擇性平板。基礎配方，補充物分段依方法添加。
原理
The medium derives from the Listeria enrichment broth of Lovett and co-workers and is specified in the FDA Bacteriological Analytical Manual for the enrichment of Listeria from foods.¹ʼ² Pancreatic digest of casein, papaic digest of soybean meal, and yeast extract supply nitrogen, amino acids, vitamins, and growth factors, and dextrose provides the carbon and energy source. The high phosphate content gives strong buffering capacity, preventing the fall in pH that occurs in less buffered enrichment broths and that inhibits Listeria. Sodium pyruvate degrades hydrogen peroxide and assists the recovery of sub-lethally injured cells. The sample is first incubated for 4 hours without selective agents to allow injured cells to resuscitate. Acriflavine, which suppresses Gram-positive competitors, nalidixic acid, which inhibits Gram-negative bacteria, and cycloheximide or natamycin, which inhibit fungi, are then added. The medium contains no differential indicator; enrichment cultures are subcultured onto selective agar for isolation and identification.²
配製與使用
Suspend 48.06 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Dispense into suitable containers. Sterilize by autoclaving at 121°C for 15 minutes. After the sample has been incubated in the base medium at 30°C for 4 hours, aseptically add the selective supplement to give final concentrations of 10 mg/L acriflavine hydrochloride, 40 mg/L nalidixic acid, and 50 mg/L cycloheximide or 25 mg/L natamycin. Test the finished medium for performance using stable, standard control cultures.
