產品說明
Tryptose Sulfite Cycloserine Agar（胰化蛋白腖－亞硫酸鹽－環絲胺酸培養基，TSC）是產氣莢膜桿菌（Clostridium perfringens）的標準計數平板：D-環絲胺酸抑制多數雜菌，亞硫酸鈉與檸檬酸鐵銨讓還原亞硫酸鹽的梭菌長成黑色菌落，以傾注或雙層平板法厭氧培養後計數。台灣食藥署「產氣莢膜桿菌之檢驗」、ISO 7937 與 FDA BAM 都指定使用，是熟肉製品、便當與調理食品檢驗的常備項目。環絲胺酸依方法於冷卻後加入。食品級原料，Halal 認證。
原理
Tryptose Sulfite Cycloserine Agar was developed by Harmon and co-workers for the enumeration of Clostridium perfringens in foods.¹ Tryptose, soy peptone, and yeast extract supply nitrogen, amino acids, vitamins, and growth factors. Sodium metabisulphite and ferric ammonium citrate act as an indicator of sulphite reduction: sulphite-reducing clostridia reduce sulphite to sulphide, which reacts with iron to form black iron sulphide, so that colonies appear black. D-Cycloserine inhibits facultatively anaerobic competitors and restricts the size and spread of colonies, improving counting accuracy. Plates are prepared by the pour-plate technique with an overlay and incubated anaerobically. Characteristic black colonies are counted as presumptive Clostridium perfringens and confirmed by appropriate tests.²
厭氧菌與挑剔菌培養基
厭氧菌與營養需求較高的菌種需要含有還原劑、血液或浸出液成分的培養基，並配合厭氧培養環境。部分培養基需要在滅菌後添加血液、血清或抗生素等易受熱破壞的成分，添加時機與方式請依配製說明操作。
配製與使用
Suspend 42 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 45-50°C and aseptically add filter-sterilized D-cycloserine to a final concentration of 400 mg/L. Mix well and use for the pour-plate technique with an overlay of the same medium. Test the finished medium for performance using stable, standard control cultures.
