Milk Plate Count Agar（牛奶平板計數培養基）是乳與乳製品總生菌數計數的專用平板：在平板計數培養基的配方上加入脫脂奶粉，更貼近乳品中微生物的營養需求，讓生乳、殺菌乳與奶粉中的菌落長得更完整。ISO 4833 乳品相關方法與英國乳品標準採用，乳品廠日常品管與原料乳驗收常備。食品級原料製造。
原理
Milk Plate Count Agar is Plate Count Agar supplemented with skimmed milk powder, specified for the colony count of microorganisms in milk and milk products.¹ Tryptone supplies nitrogen, amino acids, and peptides, yeast extract provides B-group vitamins and growth factors, and glucose serves as the carbon and energy source. The skimmed milk powder contributes lactose, casein, and other milk constituents that improve the recovery of organisms adapted to the dairy environment. The skimmed milk powder is certified free of antibiotic residues; residual antibiotics from treated animals would inhibit sensitive organisms and lead to falsely low counts. The medium contains no selective agents and supports a broad range of aerobic mesophilic bacteria.²
通用與計數培養基怎麼選
平板計數培養基用於食品、飲用水與環境檢體的總生菌數計數；營養培養基與胰化大豆培養基是非挑剔性細菌的通用選擇，後者營養較豐富，也用於無菌試驗與環境監控；腦心浸出液培養基營養最豐富，適合鏈球菌等營養需求較高的菌種。需要計數就用瓊脂版，需要增殖就用液體版。
配製與使用
Suspend 19.5 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Avoid overheating. Cool to 45-50°C and use for the pour plate technique. Test the finished medium for performance using stable, standard control cultures.
