產品說明
Urea Agar Base (Christensen)（克氏尿素培養基基礎）是尿素酶試驗的斜面培養基：培養基滅菌後加入過濾除菌的尿素溶液，具尿素酶的菌株把尿素分解成氨，pH 上升使酚紅轉成桃紅色。變形桿菌數小時內強陽性，克雷伯氏菌較慢，沙門氏菌與大腸桿菌陰性，是分離平板可疑菌落排除變形桿菌的常用一步。基礎配方，尿素依說明另加。食品級原料，Halal 認證。
原理
Urea Agar was described by Christensen to detect urease activity in a wider range of organisms than was possible with earlier, more strongly buffered urea media.¹ Peptone supplies nitrogen and growth factors, and glucose provides an energy source that supports the growth of organisms with weak urease activity. Sodium chloride maintains the osmotic equilibrium, and the low concentration of phosphate buffer allows small changes in pH to be detected. Organisms that produce urease hydrolyse urea to ammonia, which raises the pH and changes the phenol red indicator from yellow-orange to pink-red. Strong urease producers such as Proteus species give a positive reaction within a few hours, while weaker producers such as Klebsiella species may require longer incubation. Salmonella does not produce urease, and the medium is used in the biochemical confirmation of Salmonella.²
生化鑑定培養基的用途
生化鑑定培養基用來觀察菌株對特定基質的利用能力，藉由顏色變化、產氣或產生澄清環判讀結果，是傳統菌種鑑定與品管確認的基礎工具。每一種培養基對應一項生化特性，通常需要搭配數種一起判讀，並以已知菌株作為陽性與陰性對照。
配製與使用
Suspend 24.01 g of powder in 950 mL of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 45-50°C and aseptically add 50 mL of a filter-sterilized 40% urea solution. Mix well, dispense into sterile tubes, and allow to set in a slanted position. Test the finished medium for performance using stable, standard control cultures.
