產品說明
LB Broth Miller 是分子生物學與基因工程實驗室最通用的大腸桿菌液體培養基。胰蛋白腖提供胺基酸與胜肽，酵母抽出物補足 B 群維生素與核苷酸，氯化鈉維持滲透壓平衡，適用於質體增殖、勝任細胞製備與重組蛋白表現前的菌體擴增。Miller 配方的氯化鈉含量高於 Lennox 配方，是多數分子選殖操作預設使用的版本。
原理
LB Broth Miller is a nutrient-rich medium widely used for the cultivation of Escherichia coli in molecular biology and recombinant DNA work.¹ Tryptone provides nitrogen, amino acids, and peptides, while yeast extract supplies B-complex vitamins, nucleotides, and additional nitrogen and carbon sources. Sodium chloride maintains the osmotic balance and provides sodium ions for membrane transport. The Miller formulation contains 10 g/L of sodium chloride, a higher concentration than the Lennox and Luria formulations, making it suitable for applications requiring higher ionic strength.² The medium supports rapid, high-density growth and is routinely used for the propagation of plasmid-bearing strains and the preparation of competent cells.
Miller、Lennox 與低鹽配方怎麼選

配製與使用
Suspend 25.0 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121 °C for 15 minutes. Test the finished medium for performance using stable, standard control cultures.

食品級與實驗室級的差別
市面上多數培養基供應商使用實驗室級（laboratory grade）原料，在重金屬殘留、微生物污染管控與原料來源追溯上的標準較低，用於純研究足夠，但不適合對原料品質有更高要求的場景。上鼎的產品以食品級（food-grade）原料製造，生產全程在通過 ISO 22000 食品安全管理系統認證的設施內進行。

常見問題
01LB Broth Miller 和 Lennox 的主要差異是什麼？
兩者最主要的差異在氯化鈉濃度，Miller 配方高於 Lennox 配方。Miller 是大多數標準大腸桿菌實驗流程的預設選擇；Lennox 適合電穿孔轉形或對高滲透壓敏感的菌株。
02做轉形實驗應該用哪一種 LB 培養基？
化學轉形法（熱休克）的回復培養步驟通常使用 LB Broth Miller 或 Lennox，37°C 培養 45 至 60 分鐘後再鋪盤。電穿孔轉形建議使用低鹽的 Lennox 配方，避免產生電弧。
03LB Broth 和 Nutrient Broth 有什麼不同？
兩者都是通用液體培養基但組成不同。LB Broth 以胰蛋白腖、酵母抽出物與氯化鈉構成，專為大腸桿菌及重組菌株最佳化，是分子生物學實驗室的標準配方；Nutrient Broth 組成較簡單，適合一般微生物培養，但不特別針對重組菌株。分子生物學與基因工程實驗建議優先選用 LB Broth。

每公升去離子水加 25 g，滅菌後 pH 6.0 ± 0.5；配製與滅菌條件依規格書。
