원리
기초 배지는 King's Medium A에서 유래하였으며, 질소, 아미노산 및 성장인자의 공급원으로 젤라틴 펩톤과 카제인 가수분해물을, 탄소원 및 에너지원으로 글리세롤을 함유합니다. 황산칼륨과 염화마그네슘은 Pseudomonas aeruginosa의 색소 생성을 촉진합니다. CFC 첨가제를 사용하면 세트리마이드, 푸시딘산 및 세팔로틴이 육류 및 유제품의 경쟁 세균총을 억제하여, 냉장 식품의 주요 부패균인 Pseudomonas 속을 선택적으로 계수할 수 있습니다.¹ CN 첨가제를 사용하면 고농도의 세트리마이드와 날리딕스산에 의해 Pseudomonas aeruginosa에 선택적인 배지가 되며, 막여과법으로 물속의 이 균을 계수합니다.² 청록색 색소인 피오시아닌을 생성하는 집락은 Pseudomonas aeruginosa로 확정하며, 피오시아닌을 생성하지 않는 형광 집락은 추가 확인이 필요합니다. 이 배지는 물에 대해 ISO 16266, 육류에 대해 ISO 13720, 우유 및 유제품에 대해 ISO/TS 11059에 규정되어 있습니다.³
조제 방법
Suspend 48.4 g of powder in 1 L of purified water containing 10 mL of glycerol. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 45-50°C and aseptically add the CFC selective supplement for Pseudomonas species, or the CN selective supplement for Pseudomonas aeruginosa. Mix well and pour into sterile Petri plates. Test the finished medium for performance using stable, standard control cultures.
