Legionella CYE Agar Base(활성탄 효모 추출물 한천배지, CYE/BCYE)는 Legionella pneumophila 분리용 기초 배지입니다. 효모 추출물이 성장인자를 공급하고, 활성탄이 배지 중 Legionella에 유독한 과산화물과 자유 라디칼을 흡착하며, 사용 직전에 ACES 완충제, L-시스테인 및 피로인산제이철 보충물을 첨가하여 BCYE를 완성하고, 선택성이 필요한 경우 항생제를 첨가합니다. 냉각탑, 온수 시스템 및 병원 용수에 대해 ISO 11731에 규정되어 있습니다. 기초 조성이며, 보충물은 시험법에 따라 첨가합니다.
원리
Charcoal Yeast Extract Agar는 Legionella pneumophila의 1차 분리 배지로 Feeley 등이 개발하였으며, 이후 ACES 완충제와 α-케토글루타르산을 첨가하여 개량되었습니다.¹ʼ² 효모 추출물이 질소, 비타민 및 성장인자를 공급합니다. 활성탄은 억제 물질을 흡착하고 조제 및 배양 중 생성되는 과산화수소와 기타 독성 산소 유도체를 분해하여 배지를 해독합니다. 성장 보충물은 Legionella에 특이적으로 필요한 성분, 즉 이 속의 필수 아미노산인 L-시스테인과 가용성 철원인 피로인산제이철을 공급합니다. 조제 방법
Suspend 25 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 45-50°C and aseptically add the reconstituted BCYE growth supplement and, for contaminated samples, the selective supplement. Mix well and pour into sterile Petri plates, swirling continuously to keep the charcoal in suspension. Test the finished medium for performance using stable, standard control cultures.
