Cleanroom and ultrapure water microbial monitoring: settle plates, surfaces and water systems
Standard media for ISO 14698, USP <1231> and USP <61> monitoring

THE SITUATION AND THE PAIN POINTS
Fabs, panel plants and their material suppliers (specialty chemicals, electronic gases, photoresists, packaging) all monitor cleanrooms and water systems: settle plates and surface contact culture, heterotrophic counts of ultrapure and cooling water, and microbial limits on raw materials and products. The volumes are large and the schedules fixed, so media must be of fixed formulation and consistent between batches, with versions that neutralise disinfectant residues.
OUR APPROACH
For settle plates and surfaces: Tryptic Soy Agar and TSA with lecithin and polysorbate (to neutralise disinfectant residues), with Sabouraud Dextrose Agar for mould monitoring. For heterotrophic counts of ultrapure and cooling water: R-2A Agar and R-2A Broth (low-nutrient formulations for the slow growers of purified water), plus Plate Count Agar and Yeast Extract Agar for general counts. For microbial limit tests: Fluid Thioglycollate Medium, Buffered Sodium Chloride-Peptone Solution, Letheen and TAT Broth. All made to standard formulations with a COA and cultural response data for every batch.
- R-2A low-nutrient formulation, the standard for slow growers in purified water
- TSA and Letheen with neutralisers, so disinfected surfaces can still be counted
- Aligned with ISO 14698, USP <1231>, USP <61>/<62> and EP 2.6.12
- 500 g to 25 kg packs with stock on hand for fixed monitoring schedules

Frequently asked questions
01Why use R-2A for heterotrophic counts of ultrapure water?
Organisms in purified water live in a very low-nutrient environment and often fail to grow on rich media. R-2A is a low-nutrient formulation that, with lower temperatures and longer incubation (for example 20 to 28 °C for 5 to 7 days), gives higher and more consistent counts than PCA, the approach adopted by USP <1231> and ISO 6222.
02Do settle plate media need neutralisers?
Cleanrooms are wiped with disinfectants regularly and residues inhibit growth on plates. Use TSA with lecithin and polysorbate for contact plates and post-disinfection monitoring; plain TSA is adequate for routine air settle plates.
03Can cultural response data be provided for audits?
Yes. Each batch COA includes cultural response results against reference strains, and the specification sheet lists the formulation and test strains; both are downloadable after member login.
