HEK293 and insect cells: transient expression and viral vectors
Hydrolysate supplementation after transfection to raise transient expression

THE SITUATION AND THE PAIN POINTS
Research-stage recombinant proteins, antibody fragments and viral vectors are often made by HEK293 transient expression or insect-cell baculovirus systems. Transfected cells are under heavy load and expression is often inconsistent; many labs use casein-derived Tryptone N1 and want a plant-derived option of comparable performance.
OUR APPROACH
A public report from Organotechnie R&D expressed IL-4 in HEK293-6E and compared several plant-derived C-Cell grades with Tryptone N1; pea P112, guar G115 and soy S204 gave markedly higher expression than the control. For insect cells, yeast-derived Y109 and Y121 apply. Tryptone N1 is also available for labs staying with casein.
- P112, G115 and S204 outperformed Tryptone N1 in a published study
- Plant-derived, animal-origin-free
- Yeast-derived Y109 and Y121 for Sf9 and other insect cells
- Samples with a trial protocol for direct comparison in your expression system

Frequently asked questions
01When should it be added, before or after transfection?
In the published study it was added to the medium after transfection at 0.5% (5 g per litre). Add after transfection and compare 1, 2.5 and 5 g/L.
02Can it replace Tryptone N1 directly?
Swap at the same concentration and compare. In the published study P112, G115 and S204 gave higher expression, but each construct and cell line differs, so test with samples.
