المبدأ
The medium is the formulation recommended by the American Type Culture Collection for the cultivation of Escherichia coli K-12 and its derivatives.¹ Tryptone and yeast extract supply nitrogen, amino acids, vitamins, and growth factors, dextrose provides a readily fermentable carbon and energy source, and sodium chloride maintains the osmotic equilibrium. Calcium chloride supplies calcium ions, and thiamine supports the growth of thiamine-requiring K-12 derivatives commonly used in laboratory work. The heat-sensitive components are supplied separately and added after sterilisation.
طريقة التحضير
Suspend 19.0 g of Part A in 950 mL of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Dissolve 1.304 g of Part B in 50 mL of purified water and sterilize by filtration. Cool Part A and aseptically add the 50 mL of Part B. Mix well and dispense into sterile tubes or flasks. Test the finished medium for performance using stable, standard control cultures.
