Biological Treatment of Semiconductor Wastewater: Ammonia, TMAH and the Role of Culture Media

What is in fab wastewater

Semiconductor processes consume large volumes of ultrapure water and the wastewater is segregated by source. Three streams matter for microbiology:

  • Ammonia nitrogen: wafer cleaning with ammonia (SC-1 and similar), chemical mechanical polishing (CMP) and some etch steps produce ammonia-bearing wastewater. Discharge limits apply, and chemical options (air stripping, breakpoint chlorination) are costly or create by-products.
  • TMAH (tetramethylammonium hydroxide): the main component of photoresist developer, used in large volumes and highly toxic, the most scrutinised component of semiconductor wastewater in Taiwan.
  • Organics: isopropanol, cleaning agents and organic additives in slurries, expressed as COD.

All three can be treated biologically at lower cost than chemical methods, so the wastewater plants of large fabs and panel plants generally include biological units, typically membrane bioreactors (MBR) and fixed- or moving-bed biofilters.

Who eats what

Component Microorganisms Action
Ammonia Nitrifiers (Nitrosomonas, Nitrobacter and others), anammox bacteria Ammonia to nitrite and nitrate, then to nitrogen gas by denitrifiers or anammox
TMAH Acclimated mixed cultures or isolates (Pseudomonas, Hyphomicrobium, Methylobacterium and others in the literature) Degraded to trimethylamine and dimethylamine, finally mineralised to ammonia and carbon dioxide
Organics (IPA etc.) Heterotrophs of ordinary activated sludge Oxidised to carbon dioxide and water

Nitrifiers are autotrophs, slow growing and sensitive to pH and temperature; a biological tank takes weeks to build enough biomass. TMAH degraders must be acclimated with gradually increasing concentrations. This is why wastewater plants buy bioaugmentation powders or liquid cultures to seed and re-seed their systems.

Where culture media and nitrogen sources come in

The tank itself feeds on wastewater and needs no medium. Media and nitrogen sources are used upstream and around the tank:

  1. Scaling up bioaugmentation products: suppliers grow isolated or acclimated strains in fermenters and turn them into powders or liquids. Seed culture and maintenance use standard media (Nutrient Broth, Tryptic Soy Broth); large-vessel scale-up uses industrial nitrogen sources (peptone, yeast extract, soy peptone) in kilogram to tonne quantities.
  2. Start-up and acclimation of biological tanks: after commissioning or a tank turnover the influent C:N ratio is often wrong, and nitrogen must be added so the sludge can grow. The supplement is usually peptone or yeast extract, which is inexpensive, dissolves quickly and has a specified total nitrogen.
  3. Re-seeding after shock loads: when a process change or incident knocks the biomass down, bioaugmentation products plus nutrients bring the system back quickly.
  4. QC and research: viable counts of bioaugmentation products and microbial checks of tank sludge use Plate Count Agar, Nutrient Agar and R-2A.

The medium for nitrifiers themselves is a mineral formulation (ammonium salts, bicarbonate, phosphate, trace elements) without peptone; peptone and yeast extract serve the accompanying heterotrophs, mixed-sludge acclimation, and the carrier and protectant formulations of the products.

Selection principles

  • Standard formulations for scale-up and QC: fixed formulation and a COA for every batch, so a new batch needs no re-validation.
  • Industrial nitrogen sources for large vessels and tank feeding: dose from the total and amino nitrogen specification; 25 kg bulk packs keep cost down. Plant and yeast sources avoid the controls that animal-derived materials require.
  • Dose by C:N ratio: calculate from influent COD and the target ratio (roughly 100:5:1 C:N:P for ordinary heterotrophic sludge), then convert to kilograms using the total-nitrogen percentage on the specification sheet.
  • Stock on hand: re-seeding needs are usually urgent, so the supplier must hold inventory.

The other job in the same supply chain: cleanroom and water monitoring

Fabs and their material suppliers also monitor microbiology: settle plates and surface contact culture in cleanrooms (TSA, TSA with neutralisers, SDA), heterotrophic counts of ultrapure and cooling water (R-2A per USP <1231> and ISO 6222), and microbial limit tests on raw materials and products (USP <61>/<62>). These all use standard media; see the application page on cleanroom and ultrapure water monitoring.

Frequently asked questions

How low can biological treatment take TMAH? Literature and plant data show acclimated systems reducing several hundred mg/L of TMAH to single-digit mg/L or below, but influent concentration, retention time and temperature matter greatly; each plant's design and measured performance govern.

Does STBIO MEDIA sell nitrifiers or TMAH degraders? No. We supply the media and nitrogen sources for scale-up and acclimation; the strains are kept by bioaugmentation suppliers or the plants themselves.

Can you help with formulations? Yes. Tell us the strain type, fermentation volume and target cell count and we can suggest a starting seed medium and nitrogen source with dosing.
