C-Cell sample trial protocol
Follow this page once your C-Cell samples arrive. In two to three weeks you will know which grade, at what dose, keeps your cells growing at a lower serum level. Send us the data and we will help interpret it.
1. Which grades to test
| CHO | S104 · P112 · Y109 | The manufacturer validates mainly on CHO |
| HEK293 | P112 · G115 · S204 | Top three expressers in a published study |
| Vero | W106 · S104 · P112 | Published Vero serum-reduction study |
| MSC / EVs | P112 · S104 | Published MSC extracellular vesicle study |
| Sf9 | Y109 · Y121 | Manufacturer data indicate insect-cell use |
2. Prepare a stock
- Dissolve 30 g per litre (3% w/v) directly in the culture medium you will test, i.e. 3 g per 100 mL, and stir until fully dissolved.
- Sterilise through a 0.2 µm filter.
- Dilute stepwise in the same medium to the test concentrations.
- Prepare the stock on the day of use.
3. Dose: adherent and suspension cells differ tenfold
In published studies, adherent Vero cells used 0.5 g per litre while suspension HEK293 used 5 g per litre. Too high a dose may not help adherent cells, so pick three levels by culture mode:
| Low | Mid | High | |
|---|---|---|---|
| Adherent (Vero, MSC) | 0.25 g/L | 0.5 g/L | 1 g/L |
| Suspension (HEK293, CHO, Sf9) | 1 g/L | 2.5 g/L | 5 g/L |
4. Groups
| A | Current level (e.g. 10%) | None |
| B | Reduced (e.g. 5%) | None |
| C · D · E | Same as B | Low, mid and high |
Run C, D and E for each C-Cell grade, at least in triplicate. To see how low you can go, add a further serum level (e.g. 1%). If you already run serum-free, B is your current formulation and C to E add C-Cell to it.
5. Culture and records
- Keep seeding density, feeding and harvest time as usual; change only serum and C-Cell.
- For long-term stability, passage three to five times and record the doubling time of each passage.
- Record viable cell density and viability, doubling time, product yield (antibody, recombinant protein, viral titre or EV count) and cell morphology.
6. Send us the results
Put each group's data in one table and email it to [email protected]. We will help interpret it and recommend the grade and working dose.
