Overview
Tryptic Soy Broth H is the Halal-certified version of TSB, identical in formulation and use: tryptone and soy peptone supply nitrogen, glucose the carbon source, and phosphate buffers the pH, supporting fastidious and non-fastidious bacteria alike. It serves routine culture, enrichment, sterility testing and environmental monitoring, with growth performance verified against reference strains before release. Made from food-grade raw materials with Halal documentation for pharmaceutical, food and contract customers who need it.
Principle
Tryptic Soy Broth H, described in the pharmacopoeias as Soybean-Casein Digest Medium, is a general-purpose medium that supports the growth of a wide range of aerobic and facultatively anaerobic organisms, including many fastidious species.¹ Pancreatic digest of casein and papaic digest of soybean meal together provide nitrogen, amino acids, peptides, and the natural growth factors required by fastidious organisms; the soybean digest is particularly rich in carbohydrates. Sodium chloride maintains the osmotic equilibrium of the medium, dipotassium hydrogen phosphate acts as a buffer to limit pH shifts during growth, and dextrose serves as the carbon and energy source. The absence of selective or inhibitory agents makes the medium suitable for sterility testing, microbial limit testing, environmental monitoring, and the general propagation and maintenance of cultures.²
Choosing a general-purpose or enumeration medium
Plate Count Agar is used for total viable counts in food, water, and environmental samples. Nutrient Broth and Tryptic Soy media are the general-purpose choices for non-fastidious bacteria, with the latter richer and also used for sterility testing and environmental monitoring. Brain Heart Infusion is the richest of the three and suits fastidious organisms such as streptococci. Use the agar form for enumeration and the broth for propagation.
Directions
Suspend 30 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Dispense into tubes or flasks. Sterilize by autoclaving at 121°C for 15 minutes. Avoid overheating to prevent caramelization of the dextrose. Test the finished medium for performance using stable, standard control cultures.
