Overview

OGYE Agar Base (oxytetracycline glucose yeast extract agar) is a selective count plate for yeasts and moulds in food: yeast extract and glucose support fungal growth, oxytetracycline added before use suppresses bacteria, and the neutral pH lets injured yeasts recover better than on acidified media. Adopted by UK public health laboratories and many food methods as an alternative to acidified PDA. A base formulation; add oxytetracycline per the method.

Principle

OGYE Agar was described by Mossel and co-workers for the selective enumeration of yeasts and moulds in foods.¹ Yeast extract supplies B-group vitamins, nitrogen, and growth factors, and glucose provides the carbon and energy source. Unlike media that rely on acidification to suppress bacteria, OGYE Agar is used at a near-neutral pH, which avoids acid injury and improves the recovery of stressed yeasts and moulds from processed foods. Selectivity is provided instead by oxytetracycline, which inhibits a broad range of bacteria. For heavily contaminated samples, gentamicin may be added in addition to oxytetracycline to suppress resistant bacteria.²

Directions

Suspend 37 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Avoid overheating. Cool to 45-50°C and aseptically add filter-sterilized oxytetracycline to give a final concentration of 100 mg/L. Mix well and pour into sterile Petri plates. Test the finished medium for performance using stable, standard control cultures.
