Overview
Principle
M-Broth was developed by Sperber and Deibel for the rapid detection of Salmonella in dried foods and feeds using broth cultures and serological reactions.¹ Yeast extract and pancreatic digest of casein supply nitrogen, amino acids, vitamins, and growth factors, and D-mannose serves as the carbon and energy source. Dipotassium hydrogen phosphate buffers the medium, and sodium chloride maintains the osmotic equilibrium. Manganese, magnesium, and iron supply essential ions, and polysorbate 80 provides fatty acids that stimulate growth. The medium supports rapid multiplication of Salmonella and the development of flagella, enhancing the expression of the flagellar antigens detected in immunoassays. It is used for a short post-enrichment step, typically 6 to 8 hours, between selective enrichment and immunological detection.²
Directions
Suspend 36.23 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Dispense into tubes. Sterilize by autoclaving at 121°C for 15 minutes. Test the finished medium for performance using stable, standard control cultures.
