Overview

Principle

Nitrogen-fixing organisms are free-living bacteria that grow on a medium containing no source of combined nitrogen, deriving their nitrogen requirement from atmospheric nitrogen gas.¹ The nitrogen assimilated into cell protein is subsequently mineralized in the soil, contributing to the nitrogen available to crop plants. Jensen's Medium is formulated according to Jensen and contains no nitrogen source, so that only nitrogen-fixing organisms are able to grow.¹ Sucrose provides the carbon and energy source. Sodium molybdate enhances nitrogen fixation, as molybdenum is an essential cofactor of the nitrogenase enzyme complex. Dipotassium phosphate buffers the medium, magnesium sulphate and ferrous sulphate supply essential ions, and sodium chloride maintains the osmotic equilibrium. Calcium carbonate neutralizes the acids produced during metabolism and renders the medium slightly opalescent.

Directions

Suspend 39.1 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 45-50°C. Mix well to evenly suspend the calcium carbonate and pour into sterile Petri plates. Test the finished medium for performance using stable, standard control cultures.
