Overview
Motility Nitrate Medium, Buffered is the semi-solid medium for confirming Clostridium perfringens: after stab inoculation, the non-motile C. perfringens grows only along the stab line, and a red colour on adding nitrate reagents shows reduction of nitrate to nitrite; the two results together separate it from other clostridia. Specified in the confirmation steps of the Taiwan FDA C. perfringens method, the FDA BAM and ISO 7937, and read alongside lactose-gelatin medium.
Principle
Peptone and enzymatic hydrolysate of animal tissue supply amino acids and other complex nitrogenous substances. Agar is added to obtain a semisolid gel that demonstrates motility of the organism along the stab line of inoculation; growth of motile organisms extends out from the line of inoculation, while non-motile organisms grow only in the inoculated area. The medium contains glycerol and galactose to improve the consistency of the nitrate reduction reaction across different strains. In the nitrate reduction test, a pink to red color develops after addition of the reagents if nitrite is present, indicating that nitrate reduction has occurred. Some organisms further reduce nitrite to ammonia, which can be detected by adding a small amount of zinc dust to tubes showing no color.
What biochemical identification media are for
Biochemical identification media reveal whether a strain can use a particular substrate, read through colour change, gas production, or a zone of clearing. They remain the basis of conventional identification and confirmatory quality control. Each medium covers one characteristic, so several are normally read together alongside known positive and negative control strains.
Directions
Suspend 19.5 g of powder in 1 L of purified water containing 5 mL of glycerol. Heat with continuous stirring and bring to the boil for 1 minute. Dispense into test tubes to make them half full. Sterilize by autoclaving at 121°C for 15 minutes. Test the finished medium for performance using stable, standard control cultures.
