Overview
Starch Agar is an agar (solid) medium, a biochemical identification medium read by colour change or gas production. Starch hydrolysis is read with iodine. STBIO manufactures it from food-grade raw materials in an ISO 22000 certified facility, with a COA for every batch. This is the Halal-certified version.
Principle
Enzymatic hydrolysate of animal tissue and beef extract provide nitrogen, vitamins, amino acids, and other nutrients. Soluble starch is the substrate for the detection of amylase activity. Sodium chloride maintains the osmotic equilibrium. Organisms that produce the extracellular enzyme amylase hydrolyze starch in the medium. After incubation, the surface of the agar is flooded with Gram's iodine solution. Starch hydrolysis is indicated by a colorless zone surrounding the colonies, while a blue or purple zone indicates that starch has not been hydrolyzed. The size of the clear zone is proportional to the starch-hydrolyzing activity of the organism.
What biochemical identification media are for
Biochemical identification media reveal whether a strain can use a particular substrate, read through colour change, gas production, or a zone of clearing. They remain the basis of conventional identification and confirmatory quality control. Each medium covers one characteristic, so several are normally read together alongside known positive and negative control strains.
Directions
Suspend 42 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Mix well and pour into sterile Petri dishes. Test the finished medium for performance using stable, standard control cultures.
