Overview
Skim Milk Agar is an agar (solid) medium, a biochemical identification medium read by colour change or gas production. Skim milk reveals casein hydrolysis (protease activity). STBIO manufactures it from food-grade raw materials in an ISO 22000 certified facility, with a COA for every batch. This is the Halal-certified version.
Principle
Casein enzymatic hydrolysate provides amino acids and complex nitrogenous compounds. Skim milk powder provides casein as the substrate and gives the medium its opacity. Yeast extract provides vitamins, and dextrose provides the carbon and energy source. Proteolytic organisms produce caseinase, which hydrolyzes casein into soluble nitrogenous compounds, indicated by a clear zone surrounding the colonies. The clearing is more pronounced when the organism also produces acid from fermentable carbohydrates in the medium.
What biochemical identification media are for
Biochemical identification media reveal whether a strain can use a particular substrate, read through colour change, gas production, or a zone of clearing. They remain the basis of conventional identification and confirmatory quality control. Each medium covers one characteristic, so several are normally read together alongside known positive and negative control strains.
Directions
Suspend 51.5 g of powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Mix well and pour into sterile Petri dishes. Test the finished medium for performance using stable, standard control cultures.
