Overview
Mueller Hinton Agar is the international standard medium for antimicrobial susceptibility testing by disc diffusion, as specified by both CLSI and EUCAST. The formulation deliberately avoids components that interfere with antibiotics, starch adsorbs toxic metabolites, and the agar content gives uniform antibiotic diffusion so inhibition zones can be read against standard tables. Used for aerobic and facultative anaerobic bacteria, with performance verified against CLSI QC strains before release.
Principle
Acid hydrolysate of casein and beef extract supply amino acids and other nitrogenous substances, minerals and other nutrients to support the growth of microorganisms. Starch acts as a protective colloid against toxic substances that may be present in the medium. Hydrolysis of starch during autoclaving provides a small amount of dextrose, which is a source of energy. Agar is the solidifying agent.
Media for anaerobes and fastidious organisms
Anaerobes and fastidious organisms require media containing reducing agents, blood, or infusion components, together with an anaerobic incubation environment. Some formulations need heat-labile supplements such as blood, serum, or antibiotics added after sterilisation; follow the preparation instructions for the timing and method.
