Overview
Principle
The base is derived from King's Medium A and contains gelatin peptone and casein hydrolysate as sources of nitrogen, amino acids, and growth factors, with glycerol as the carbon and energy source. Potassium sulphate and magnesium chloride enhance the production of pigments by Pseudomonas aeruginosa. With the CFC supplement, cetrimide, fusidic acid, and cefalotin suppress the competing flora of meat and dairy products, allowing the selective enumeration of Pseudomonas species, which are important spoilage organisms of chilled foods.¹ With the CN supplement, the higher concentration of cetrimide together with nalidixic acid makes the medium selective for Pseudomonas aeruginosa, which is enumerated in water by membrane filtration.² Colonies producing the blue-green pigment pyocyanin are regarded as confirmed Pseudomonas aeruginosa, while fluorescent colonies without pyocyanin require further confirmation. The medium is specified in ISO 16266 for water, ISO 13720 for meat, and ISO/TS 11059 for milk and milk products.³
Directions
Suspend 48.4 g of powder in 1 L of purified water containing 10 mL of glycerol. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. Cool to 45-50°C and aseptically add the CFC selective supplement for Pseudomonas species, or the CN selective supplement for Pseudomonas aeruginosa. Mix well and pour into sterile Petri plates. Test the finished medium for performance using stable, standard control cultures.
