Overview
Marine Agar 2216 is an agar (solid) medium. It is with a salt profile close to seawater, used to isolate and cultivate marine bacteria. STBIO manufactures it from food-grade raw materials in an ISO 22000 certified facility, with a COA for every batch. This is the Halal-certified version.
Principle
Marine Agar 2216 is formulated according to the composition described by ZoBell to reproduce the major mineral content of seawater, thereby supporting the abundant growth of marine bacteria.¹ The medium contains a high concentration of sodium chloride together with magnesium, calcium, potassium, and trace minerals that approximate the ionic composition of natural seawater.² Peptone and yeast extract serve as the sources of nitrogen, carbon, amino acids, and vitamins, while ferric citrate supplies iron required by many marine organisms. Because most marine bacteria are heterotrophic and sensitive to elevated temperatures, the medium is cooled to 42°C before pour-plate inoculation to preserve viability. Both pour plate and spread plate techniques may be used for enumeration.
Directions
Suspend 55.3 g of the powder in 1 L of purified water. Heat with continuous stirring and bring to the boil for 1 minute. Sterilize by autoclaving at 121°C for 15 minutes. For the pour plate technique, cool the medium to 42°C before inoculation, as most marine bacteria are thermosensitive. Mix well and pour into sterile Petri plates. Test the finished medium for performance using stable, standard control cultures.
Cultural response
